Mustafa was born in Portland, Oregon, the youngest of seven children, to Shahidah Mustafa-Davis and John Wali Mustafa, a Muslim family. When he was about five years old, his family moved to Mission Viejo, California, where his father established a limousine service. They moved to Oxnard two years later, after the death of his father.
Mustafa got into track as a senior at Santa Clara High in Oxnard, California and was a member of the basketball team. Geof Foley, then the track coach at the school, convinced Mustafa to specialize in the decathlon, and Mustafa placed fourth at the 1992 Golden West Invitational. He apparently wanted to play football for the Saints his senior season in 1991, but remained committed to the basketball team.
After an outstanding run with the Saints basketball team, many expected a Division I school to recruit Mustafa. When no one did, Mustafa, rather than accept one of the few Division II offers he had, Mustafa decided to play at Santa Monica but quit the team soon after arriving. He transferred to Moorpark College to concentrate on track with Green, who recruited Mustafa in high school. He eventually came to join the football team at Moorpark.
In 1995, Isaiah transferred to Arizona State University, where he took History and played as a starting wide receiver for the Arizona State Sun Devils until he was benched in his senior year. He graduated in 1997. After graduating, he played four years in NFL practice squads as a wide receiver.
Mustafa's acting career began with bit roles on various television programs, such as L.A. Law, productions and films. He gained sudden fame in February 2010 for his role in the popular The Man Your Man Could Smell Like advertising campaign for Old Spice, which opened up various opportunities for him.
Isaiah signed a talent deal with NBC shortly after. He earned a guest spot on the NBC show Chuck the same year. He appeared in the film Horrible Bosses and hosted the TBS special Funniest Commercials of the Year in 2010, and again the following year. In 2011, he voiced a Gear in the video game Gears of War 3.
